The effects of selective serotonin reuptake inhibitors on extracellular 5-HT in the hippo- campus of 5-HT1B receptor knockout mice

چکیده
The effects of two selective serotonin reuptake inhibitors (SSRIs) on 5-hydroxytryptamine (5-HT) in the hippocampus were studied in wildtype and in 5-HT1B receptor knockout (KO) mice using in vivo microdialysis. Basal 5-HT levels were not different between the two genotypes. The functional absence of 5-HT1B receptors was examined in the knockout mice by local infusion of the 5-HT1B receptor agonist, CP93129 (1,4-Dihydro-3-(1,2,3,6-tetrahydro-4-pyridinyl)-5Hpyrrolo[3,2-b]pyridin-5-one) into the hippocampus. CP93129 (1 μM) decreased 5HT levels in wildtype mice, but not in 5-HT1B KO mice. Systemic administration of the SSRI paroxetine (5 mg/kg i.p.) increased extracellular 5-HT levels. The increase of 5-HT in 5-HT1B KO mice was almost twofold higher than in wildtype mice. Systemic administration of SSRIs stimulates both terminal 5-HT1B autoreceptors and somatodendritic 5-HT1A autoreceptors. Therefore, another

Extracellular serotonin in the prefrontal cortex is limited through terminal 5-HT1B autoreceptors
Rationale: 5-HT autoreceptors regulate extracellular 5-HT levels and have been suggested to limit the effects of acute treatment with selective serotonin reuptake inhibitors (SSRI). Objectives: The role of terminal 5-HT1B autoreceptors was assessed by comparing the effects of a SSRI on extracellular 5-HT in wildtype and 5-HT1B receptor knockout mice, and by using a 5-HT1B receptor antagonist. S...
متن کاملInteractions between serotonin and dopamine in the striatum are not mediated by striatal 5-HT1B receptors
In vivo microdialysis was performed in wildtype and 5-HT1B receptor knockout mice to explore the role of 5-HT1B receptors in striatal serotonin (5-HT) and dopamine (DA) outflow. Local administration of the selective 5-HT reuptake inhibitor fluvoxamine (0.1 – 10 μM) dose-dependently increased 5-HT to the same extent in wildtype and 5-HT1B receptor knockout mice. The 5-HT1B receptor agonist CP931...
متن کاملShort communication Improved efficacy of f luoxetine in increasing hippocampal 5-hydroxytryptamine outf low in 5-HT1B receptor knock-out mice
To test for the contribution of the 5-HT1B receptor subtype in mediating the effects of fluoxetine, a selective serotonin reuptake inhibitor (SSRI), we used intracerebral in vivo microdialysis in awake, freely moving 5-HT1B receptor knock-out mice. We show that a single systemic administration of fluoxetine (1, 5 or 10 mg/kg, i.p.) increased extracellular serotonin levels [5-HT]ext in the ventr...
متن کاملSerotonin1B heteroreceptor activation induces an antidepressant-like effect in mice with an alteration of the serotonergic system.
OBJECTIVE We sought to demonstrate whether the specific activation of serotonin1B (5-HT1B) heteroreceptors by systemic or local administration of the selective 5-HT1B receptor agonist anpirtoline could mediate antidepressant-like effects in mice. METHODS We confirmed the selectivity of action of anpirtoline in the forced swim test (FST) in 5-HT1B knockout mice. We then evaluated the behaviour...
متن کاملGenetic regulation of extracellular serotonin by 5-hydroxytryptamine(1A) and 5-hydroxytryptamine(1B) autoreceptors in different brain regions of the mouse.
The regulation of extracellular levels of 5-hydroxytryptamine (serotonin) (5-HT) in the striatum and ventral hippocampus was studied using in vivo microdialysis in awake, unrestrained wild-type 5-HT(1A) and 5-HT(1B) receptor knockout mice. Systemic administration of the selective serotonin reuptake inhibitor fluoxetine evoked a significant dose-dependent increase in extracellular 5-HT in both t...
